Bioindicators for Bacillus stearophilus (wet)


1. This product is a biological indicator of pressure steam sterilization (biological indicator of Bacillus stevia). For quality control, refer to USP31 (USP 31) and ISO-11138 (International Organization for Standardization). The strain used is Bacillus subtilis ATCC 35021.


2 If the sterilization treatment is incomplete, the remaining lipobacillus spores will multiply and metabolize into acid when it encounters the culture solution (containing ph indicator), the pH value will decrease, and the color of the culture solution will change from purple-red to yellow, if the sterilization treatment is complete. All the spores died, and the color of the culture medium remained purple-red.


3. There is a specially processed hydrophobic filter paper in the lid of the product. The filter paper can pass through the steam and gas without passing microorganisms, which is completely suitable for pressure steam sterilization.


4. The label on the surface of this product has the property of chemical indication, which is convenient to confirm whether it has been sterilized.



Instructions


1 In the blank space on the surface of the ACE test, use an oil-based pen to record the necessary items for sterilization management. (Such as: date, location, etc.)


2 A few branches of ACE test are placed horizontally in the sterilizer which is not easily sterilized completely.


3 After sterilization, take out the ACE test, confirm that the chemical indicator on the label turns black, and leave it to cool to room temperature for more than 15 minutes.


4 Place the completely cooled ACE test vertically into the clamp until the bottom of the plastic tube is completely stable. Press the upper part of the clamp to break the internal glass tube. At this stage, the paper that adsorbs the spores contacts the culture solution. The culture preparation is complete. Whether the slices are immersed in the culture medium and cultured in time.


5Cultivate the ACE test vertically at a temperature between 35 and 39 ° C. For comparison, please use the same batch of unused ACE test, and then culture the glass tube after breaking.


6 After confirming the sterilization effect, please remove the label and stick it in the record book.



Result judgment


1 After 48 hours of incubation, the color of the culture solution changes from fuchsia to yellow, indicating that the sterilization is incomplete. Please sterilize again.


2 If the color of the culture solution remains purple, please continue to culture for 2 days. If there is no sign of discoloration, you can judge that the sterilization is complete.


3 The biological test result is only valid if the control tube is positive. The color of the culture solution of the ACE test used for the control must be changed from purple to yellow.

1 Before use, please confirm the integrity of the product and use it within the validity period, the validity period is 24 months.


2 Biological indicators should be kept away from sterilizers and chemical disinfectants.


3 Biological indicators that have been judged to be incompletely sterilized or have expired, please discard them after sterilization.


4 After confirming the propagation of the bacteria (yellow), please do not continue the culture.
